Experimental Autoimmune Encephalomyelitis (EAE) Model

Sichuan Greentech Bioscience provides a well-established experimental autoimmune encephalomyelitis (EAE) model, which has been validated through multiple approaches. All animals are housed in AAALAC-accredited animal facilities, and in vivo pharmacodynamic evaluations are conducted in accordance with GLP management standards.


Our Experimental Autoimmune Encephalomyelitis Model

Model Induction: Experimental autoimmune encephalomyelitis (EAE) is an autoimmune demyelinating disease induced by myelin autoantigens and is widely used as an animal model of multiple sclerosis (MS). EAE shares key pathological features with MS, including inflammatory cell infiltration and demyelination within the white matter of the central nervous system (CNS). Proteolipid protein (PLP) is a major myelin protein of the CNS, and the PLP₁₃₉–₁₅₁  peptide represents an immunodominant encephalitogenic epitope that can reliably induce CNS demyelinating lesions in susceptible mice.

Animals: SPF SJL/J mice, female, 8–10 weeks old. Immunization of SJL/J mice with PLP₁₃₉–₁₅₁  can induce both acute and relapsing-remitting EAE, making this model particularly relevant for studies of MS and its relapsing-remitting disease course.

Model Induction Procedure:

① PLP₁₃₉–₁₅₁ solution (0.5 mg/mL) is mixed with an equal volume of complete Freund's adjuvant (CFA) containing 2 mg/mL Mycobacterium tuberculosis and emulsified using a dispersing homogenizer to obtain a water-in-oil emulsion.

② A total of 0.2 mL of the antigen emulsion is administered subcutaneously at two sites on either side of the abdomen (0.1 mL per site).

③ Pertussis toxin is administered intraperitoneally at 200 μL (60 ng) per animal at 0 and 48 hours after antigen emulsion administration.

Endpoints: Body weight and neurological function score (modified 15-point Weaver scoring system).
